The invention discloses a pharmaceutical composition of a 4-aminoquinoline derivative and snake venom cytotoxin-CTX1. The 4-aminoquinoline derivative is chloroquine and/or hydroxychloroquine. The pharmaceutical composition contains three ingredients A, B and C and two combinations, namely A+B and A+C, wherein A is CTX1 or other members of a CTX family; B is CQ or a medicinal derivative thereof, and C is HCQ or a medicinal derivative thereof. Compared with the prior art, the pharmaceutical composition disclosed by the invention has the advantages and significant progress that CTX1, CQ and HCQ are lysosome targeting agents respectively, low-dosage CQ or HCQ respectively enhances the effect of CTX1 on lysosome, and the dosage of CTX1 in the pharmaceutical composition is only 1/3.6-1/2.3 of individual dosage.
