The invention provides a room-temperature liquid high-concentration iodine, and a preparation method and application thereof. The liquid high-concentration iodine is in a liquid state at a temperatureof 60 DEG C or below. The preparation method comprises the following steps: (1) adding solid iodine into a reagent and/or into a medium combination formed by a reagent and a solvent, adding a catalyst, then performing stirring, performing heat preservation for reaction after the stirring, and then performing cooling to obtain a reaction solution; and (2) diluting and stirring the reaction solution obtained in the step (1), then performing standing for layering, and separating out the lower-layer liquid high-concentration iodine. According to the invention, a proper reagent is selected to react with the solid iodine under certain conditions, so that the high-concentration iodine which is in liquid state at a room temperature can be prepared.
