The invention provides a preparation method of a crystal form of N-[(S)-1-(5-(3-chloro-4-cyanophenyl)-1H-pyridine-1-yl)propyl-2-yl]-5-(1-ethoxyl)-1H-pyridine-3-formamide. The preparation method comprises the following steps: firstly, dissolving the N-[(S)-1-(5-(3-chloro-4-cyanophenyl)-1H-pyridine-1-yl)propyl-2-yl]-5-(1-ethoxyl)-1H-pyridine-3-formamide into a mixed solvent, raising the temperature,and mechanically stirring until the N-[(S)-1-(5-(3-chloro-4-cyanophenyl)-1H-pyridine-1-yl)propyl-2-yl]-5-(1-ethoxyl)-1H-pyridine-3-formamide is completely dissolved; and secondly, dropwise adding fatty alkane into a product obtained in the first step; after addition is finished, slowly reducing the temperature, mechanically stirring for 1 to 16 hours, filtering and drying a separated solid to obtain the crystal form of the N-[(S)-1-(5-(3-chloro-4-cyanophenyl)-1H-pyridine-1-yl)propyl-2-yl]-5-(1-ethoxyl)-1H-pyridine-3-formamide.
