The novel method includes steps of mixing trichoderma, aspergillus oryzae and phanerochaete chrysosporium with one another according to a proportion of 90:8:2 and carrying out solid-state fermentation; carrying out uncontrolled wet closed static fermentation for 4 days and converting the rice straw into diversified small-molecule carbohydrates; producing polyunsaturated fatty acid from straw hydrolysate by means of fermentation by the aid of fermentation trichosporon asahii at the temperatures of 25-28 DEG C; preparing the biodiesel from prepared rice straw hydrolysate and prepared unsaturated fatty acid by the aid of constructed Escherichia coli homologous recombinant bacteria with fatty acid ethyl ester (namely, biodiesel) synthesis ability.
