An Antrodia Cinnamomea spore oil and a manufacturing method thereof. The method comprises the following steps: (1) spore wall breaking: freezing Antrodia Cinnamomea asexual spores in a temperature and pressure-variable puffing apparatus to perform dehydration and puffing, then adding particles of dry ice, and grinding the powder and particle mixture with a high-energy nano-scale ball mill; (2) extraction: placing the wall-broken asexual spore powder into an extraction vessel of a supercritical CO2 extraction device, adding a co-solvent, and performing extraction at 20-60 DEG C, wherein an extraction pressure of the extraction vessel is 10-35 MPa, a CO2 flow rate is 10-40 L/h, and an extraction duration is 1-1.5 hours; after the extraction, placing into separation vessels to perform separation, wherein the pressure in a separation vessel I is 5-25 MPa, the temperature therein is 20-30 DEG C, the pressure in a separation vessel II is 7-15 MPa, and the temperature therein is 20-50 DEG C; and (3) concentration: v
