The invention relates to the technical field of EST (Expressed Sequence Tag)-SSR (Simple Sequenc Repeat) markers, in particular to a preparation method and application of a sugarcane EST-SSR marker. The method comprises the steps of screening SSR loci of a sugarcane transcriptome sequence; then utilizing nucleotide sequence designed primers at two sides of the SSR loci to obtain an EST-SSR marker primer; using the EST-SSR marker primer for marking a corresponding germplasm DNA (Deoxyribose Nucleic Acid); using capillary gel electrophoresis for verifying a marker result.
