The method comprises the steps of introducing a herbicide-resistant gene into a maintainer line B of the three-line hybrid rice through hybridization and backcrossing to obtain a maintainer line Br with the pure herbicide-resistant gene; performing hybridization by taking the maintainer line Br as a male parent and taking a herbicide-sensitive sterile line A corresponding to the maintainer line as a female parent to obtain a sterile line Ar with the herbicide-resistant gene, wherein the sterile line A is a heterozygote of the herbicide-resistant gene; and planting the sterile line Ar, and spraying a herbicide in a rice seedling stage, wherein selfed or pollen-polluted hybrid seedlings are dead, and survival plants are sterile lines with very high purity and herbicide-resistant genes.
