The invention discloses a group of peptides having a function of easing pain, and a pharmaceutical composition and application of the peptides. The peptides have amino acid sequences as shown in a sequence table SEQ ID No. 1 or a sequence table SEQ ID No. 2, or pharmaceutically acceptable salts of the peptides are provided. Based on the presence of head-end cysteine and terminal cysteine, after one or more amino acids of the amino acid sequence, as shown in the SEQ ID No. 1 or the SEQ ID No. 2, are deleted, substituted or added, polypeptides or pharmaceutically acceptable salts thereof still have the function of easing pain. The polypeptides are formed after one or more amino acids in Loop 3 of GsMTx-4 are deleted, substituted or added, and then the polypeptides having the function of easing pain are screened out from the polypeptides. According to the results of detection for a rat nociceptive pain model by the polypeptides, the inventor proves that a polypeptide short chain series having the function of eas
