Summary: | The invention relates to the production technical field of separation and preparation of complete amino acids, and provides a separation and preparation method of complete essential amino acids. The method comprises the following steps: fermenting soybean protein, which serves as a major raw material, by virtue of bacillus subtilis which is rich in protease, so that the soybean protein is hydrolyzed into free amino acids by virtue of the protease; and extracting the free amino acids with the addition of 45 times of 55% ethanol at 45 DEG C for 2.5h, wherein the leaching amount of the total free amino acids reaches 48.31mg/g or above and the yield of the total free amino acids is 55.36% of the content of the total free amino acids. The complete essential amino acids are successfully separated from the extracted total free amino acids by virtue of such methods as a similar dissolution, an isoelectric precipitation, gel filtration chromatography and the like.
