The invention discloses a detection method of fungus polysaccharide. The detection method comprises the following steps: preparing to obtain a sample; using glucose solution as a standard substance; using a sulfuric acid-anthrone or sulfuric acid-phenol method to prepare the standard curve, wherein the absorbance is ordinate and the concentration is abscissa in the standard curve; developing with the sulfuric acid-anthrone or sulfuric acid-phenol method; measuring the absorbance with a spectrophotometer to obtain the concentration of the anhydrous glucose in the corresponding sample from the standard curve according to the measured absorbance; and re-calculating to obtain the content of polysaccharide in the sample.
