Summary: | The invention discloses a preparation method of technetium-99m labeled DNA trigonal bipyramid nanoparticles. According to the preparation method, a ligand compound is reacted with an amino modified DNA oligonucleotide chain, and the product is purified to obtain a reactant A, wherein the ligand compound is a radioactive metal ligand molecule; the reactant A, the hydrochloric acid solution of stannous chloride and pertechnetate ions are mixed evenly in a solvent A, the pH value of the mixed solution is adjusted to the range of 6.0-7.5, and then a reduction coordination reaction is performed to obtain a reactant B; the reactant B is mixed evenly with the aqueous solution of the DNA trigonal bipyramid nanoparticles in a solvent B, and finally, after purification by virtue of high performance liquid chromatography, the technetium-99m labeled DNA trigonal bipyramid nanoparticles can be obtained. The preparation method is high in reaction selectivity, mild in conditions, simple to operate, relatively high in labeling yield and simple and convenient in aftertreatment steps. The technetium-99m labeled DNA trigonal bipyramid nanoparticles are good in water solubility and excellent in biocompatibility, and provide more basic information for the subsequent application of the DNA nanoparticles to an organism. |
