Summary:The invention discloses a method for extracting DNA (deoxyribonucleic acid) by virtue of the combination of a magnetic bead method and a centrifuge shield. The method comprises the following steps: (1) putting a detection material into an inner sleeve of the centrifuge shield, uniformly mixing a TES buffer solution, sodium dodecyl sarcosinate and protease K according to a volume ratio, then adding the mixture into the inner sleeve of the centrifuge shield, and putting the centrifuge shield on a metal bath to perform pyrolysis; (2) adding an adsorption solution and centrifuging again; (3) removing the inner sleeve, adding the adsorption solution, adding a magnetic bead suspension, and putting on an automatic vortex instrument to perform adsorption; (4) putting on a magnetic frame to suck away a supernatant, and then putting into a centrifuge to suck away residual liquid again; (5) adding absolute alcohol for whirling, putting on the magnetic frame to fully suck away the supernatant, and putting into the centrifuge to suck away the residual liquid again by centrifuging; (6) drying magnetic beads, adding an eluent to whirl fully, and then putting on the metal bath for incubation; and (7) performing amplification in a PCR (polymerase chain reaction) amplification solution. The method disclosed by the invention is high in detectable rate, and the final template keeping amount reaches 50-70%, so that the method is particularly suitable for detecting micro detection materials, and the field DNA detection material containing more than 100pg of templates can be detected.
