The invention discloses a stable bioactive protein or polypeptide loaded hyaluronic acid solution and a preparation method thereof. According to the solution disclosed by the invention, hyaluronic acid and/or hyaluronate are/is taken as substrate which is loaded with one or more bioactive protein or polypeptide (0-10%, W/V), and meanwhile, a hyaluronidase preparation (0-0.1%, W/V), a protein active protective agent (0-10%, W/V) and/or a penetration enhancer (0-20%, W/V) are combined. An ultra-molecular solution formed on the basis of the mutual action of positive and negative charges in a layer by layer self-assembling mode can be applied to the medical treatment of burned skin, incised skin, scalded skin and minimally invasive skin, and the solution is suitable for the external application to complete skin to take cosmetic effects of tightening and removing wrinkles.
