The method comprises the following steps: A. selecting the plateau of Allium ovalifolium on high mountain as an explant; B. peeling the plateau until reaching the diameter of 0.3-0.5cm with a sterilized scalpel, inserting into an induction medium in a polarity direction; C. cultivating at constant temperature under illumination until a clump shoot grows; D. cutting the clump shoot into a plurality of small clump shoots and transferring into a proliferation medium for multiplication of subculture; E. transferring a strong seedling into a root medium in a subculture cultivation process, opening a bottle for hardening seedling after rooting, cleaning, transferring into matrix, keeping the temperature and the humidity, and transplanting after the tissue culture seedling is survival. The acquired Allium ovalifolium has good hereditary stability, small occupied area for propagation and high propagation speed, one plateau per year can reproduce millions of Allium ovalifolium seedlings, the supplying problem of high-quality Allium ovalifolium seedlings in Allium ovalifolium planting can be effectively solved, the built production system is stable, the period is short, and the scale seedling planting requirement can be met.
