The invention provides a room-temperature vulcanized single component polyurethane structural sealant and a preparation method thereof. The sealant is prepared from the following raw materials in parts by weight based on the total weight: 100 parts of a polyurethane prepolymer, 40-80 parts of a plasticizer, 70-120 parts of filler, 0-5 parts of a thixotropic agent, 1-2 parts of an ultraviolet absorber, 1-2 parts of a light stabilizer and 0.5-1.5 parts of a catalyst. According to the polyurethane structural sealant prepared by the invention, temperature and humidity have little influence on the surface drying time and curing. Even if the sealant is applied in winter, the work beat of a customer is not over-affected, so that the defects of existing polyurethane structural sealant are overcome.
