The establishing method detects rheum emodin in the wind-dispelling and detoxifying capsules by adopting high performance liquid chromatography, wherein the condition is that the chromatographic column takes octadecyl silane bonded silica gel as a filling material; the mobile phase comprises a mobile phase A of acetonitrile and a mobile phase B of acidic aqueous solution, and gradient elute is performed; the flow velocity is 1.0mL/min; the column temperature is 30 DEG C; the detection wavelength is 210nm-400nm; and the theoretical tower plate number is not less than 8000 based on the peak of rheum emodin. The obtained fingerprint spectrum mainly refers to a chromatographic peak with main active ingredients such as the polygonum cuspidatum, fructus forsythiae, verbena and liquorice. With the adoption of the establishing method disclosed by the invention, a fingerprint spectrum which can comprehensively characterize the active ingredients of the wind-dispelling and detoxifying capsules can be effectively obtained, and the method has the characteristics of high precision, high stability and high repeatability. The obtained fingerprint spectrum can be used for ensuring the stability and consistency of the product quality, so that the product is safe and effective.
