The invention relates to calcium sulfate whisker synthetic paper and a preparation method thereof. The synthetic paper comprises the following components by mass percent: 30-50% polyolefin, 40-60% calcium sulfate whisker, 2-15% compound additives, wherein the compound additives comprise a coupling agent, a toughening agent, an antioxidant, a plasticizer and a dispersing lubricant; the preparation method comprises continuously stirring the calcium sulfate whisker, the dispersing lubricant and the coupling agent in a high-speed mixer equipped with a heating device and heating, adding the toughening agent and continuously stirring to obtain a micro-powder material, adding the micro-powder material, polyolefin, the antioxidant and the plasticizer to a twin-screw extruder unit, intermixing, plasticizing, extruding and granulating, cooling and drying the pellets to obtain the synthetic paper masterbatch, melt mixing the synthetic paper masterbatch by a single screw extruder unit and extruding a molten material solu
