Summary: | The invention discloses a construction method of an etr1 gene containing binary expression vector and an application thereof. The construction method comprises the following steps of: amplifying an Arabidopis thaliana mutant etr1 gene and cloning it onto an expression vector pEASY-T1 to obtain a pEASY-T1-etr1 plasmid, carrying out enzyme digestion on AscI and BamHI restrictive endonuclease sites of the plasmid by a double enzyme digestion system to obtain an etr1 gene fragment, carrying out double enzyme digestion on a pFGC5941 plasmid by the same double enzyme digestion system to obtain a backbone fragment, linking the etr1 gene fragment and the backbone fragment by the use of T4DNA ligase to form a circular plasmid. Therefore, the pFGC5941-etr1 binary expression system is obtained. If the system is applied in fresh cut flowers, the service life of the fresh cut flowers can be prolonged for 5-10 days. |
