A separation chamber device used for preparative free-flow electrophoresis (FFE) comprises a first synthetic glass flat plate, a second synthetic glass flat plate, an electrophoresis chamber and an electrode chamber; the electrophoresis chamber and the electrode chamber, which are formed by a separation hole and an electrode hole of an ion-exchange membrane, and a plastic film respectively, are clamped between the first and the second synthetic glass flat plates; a plastic screw passes through an upper screw hole on the first synthetic glass flat plate, a lower screw hole on the second synthetic glass flat plate and a film screw hole of the plastic film to be fixed; and an electrode buffer solution inlet and an electrode buffer solution outlet are formed on the first synthetic glass flat plate and at the positions corresponding to two ends of the electrode hole of the ion-exchange membrane and are used for circulating electrode solution. The invention can solve the problems of leak tightness and stability of
