Abstract | The invention discloses a method for screening effective shRNA of adipose triacylglycerol hydrolase gene. The method comprises the following steps of: A1, designing and synthesizing a single-chain DNA oligonucleotide segment for coded target shRNA; A2, constructing a pENTR/CMV-GFP/U6-shRNA carrier; A3, constructing a pDsRed1-C1-ATGL carrier; and A4, screening an effective sequence of the shRNA. Because an interference vector and a target gene are marked by red fluorescence and green fluorescence respectively, the process of screening interference sequences is time-saving and high in efficiency, the result is more intuitive and reliable, and a plurality of sequences can be screened at the same time. |
