The invention belongs to the technical field of medicines and discloses a water soluble cantharidin and/or norcantharidin chitosan derivative and a preparation method thereof. The derivative is obtained after reaction between the chitosan and the cantharidin or norcantharidin (norcantharidin acid and salt thereof). The chitosan derivative is shown as follows, wherein R and R' is respectively and independently selected from the cantharidin or norcantharidin (norcantharidin acid and salt thereof) and n is 0.5-3000 (the molecular weight of the chitosan is 179-1000000). The derivative has good water solubility and low toxic and side effects, and the reaction product has anti-tumor activity, alleviates the vascular stimulation of cantharidin, increases leukocyte and can be used for treating tumors.
