Summary:Disclosed are: a method for cleaving a peptide bond at the C-terminus of a peptide, wherein the method is improved in the lowering of sensitivity and produces little side reactions; and a method for determination of the C-terminal amino acid sequence of a peptide. The method for cleaving a peptide bond at the C-terminus of a peptide comprises the steps of: reacting the peptide with a solution of a perhalogenated carboxylic acid containing an alcohol so as to esterify a glutamic acid residue in the peptide; and reacting the peptide with an anhydrous alkanoic acid so as to produce a C-terminal deficient peptide wherein the C-terminal amino acid residue in the peptide is deleted one by one.
