Summary:TYK2 is a non-receptor tyrosine kinase member of the Janus kinase (JAKs) family of protein kinases. The mammalian JAK family consists of four members, TYK2, JAK1, JAK2, and JAK3. JAK proteins, including TYK2, are integral to cytokine signaling. Disorders associated with TYK2 include autoimmune disorder, inflammatory disorder, proliferative disorder, endocrine disorder, neurological disorder, and disorder associated with transplantation. The present invention provides compounds exemplified by fommla I, compositions thereof, and methods of using the same for the inhibition of TYK2, and the treatment of TYK2-mediated disorders.
