Summary:A lipase or lipase isozyme from Candida rugosa is used to stereoselectively hydrolyze esters, esterify acids or transesterify esters in aqueous/organic systems. Esters of 2-substituted acids such as 2-(4-hydroxyphenoxy) propionic acid, where the acid is not substituted at the 2position with halogen, are hydrolyzed at high enantiomeric excess in the presence of an organic solvent. A reducing agent may be present to prevent oxidation of the acid and its esters. Two lipase isozymes are purified from Candida rugosa by ion exchange chromatography or isoelectric focusing, and are used for stereoselective hydrolysis of esters, esterification of acids or transesterification of esters. The lipase or lipase isozyme may be immobilized, and immobilization is improved by the presence of polyvinyl alcohol or an organic acid such as stearic acid. The presence of a fatty acid or fatty acid ester during lipase reaction increases the rate or stereoselectivity of the reaction.
