A method of increasing the hydrodynamic size or hydrodynamic volume of a recombinant human growth hormone (hGH) polypeptide, the method comprising providing DNA encoding an hGH polypeptide having one chorionic gonadotrophin carboxy terminal peptide (CTP) unit attached to the amino terminus of said hGH and two CTP units attached to the carboxy terminus of said hGH, and expressing the recombinant CTP-modified hGH polypeptide in a host cell.
