Summary:Disclosed is a modified biotin-conjugated protein which is characterized by having any one substitution selected from the group consisting of items 1) to 6) shown below in a protein which comprises the amino acid sequence depicted in SEQ ID NO:2 or a mutant sequence thereof and has a biotin binding activity: 1) the substitution of a serine residue at position-36 in SEQ ID NO:2 for an amino acid residue incapable of forming a hydrogen bond; 2) the substitution of a tryptophan residue at position-80 in SEQ ID NO:2 for a hydrophilic amino acid residue; 3) the substitution of an aspartic acid residue at position-116 in SEQ ID NO:2 for an amino acid residue incapable of forming a hydrogen bond; 4) the substitution of a proline residue at position-46 in SEQ ID NO:2 for a threonine, serine or tyrosine residue, and the substitution of a threonine residue at position-78 in SEQ ID NO:2 for an amino acid residue incapable of forming a hydrogen bond; 5) the substitution of a proline residue at position-46 in SEQ ID NO:2 for a threonine, serine or tyrosine residue, and the substitution of an aspartic acid residue at position-116 in SEQ ID NO:2 for an amino acid residue incapable of forming a hydrogen bond; and 6) the substitution of a proline residue at position-46 in SEQ ID NO:2 for a threonine, serine or tyrosine residue, the substitution of a threonine residue at position-78 in SEQ ID NO:2 for an amino acid residue incapable of forming a hydrogen bond, and the substitution of an aspartic acid residue at position-116 in SEQ ID NO:2 for an amino acid residue incapable of forming a hydrogen bond.
