Summary: | INTRODUCTION
ESTRADIOL-17β was the third of the three ‘classical’ estrogens to be isolated from human urine (Smith et al., 1939; Huffman et al., 1940). As regards the domestic fowl, MacRae et al. (1959) isolated a small amount of crystalline estradiol-17β from the mixed excreta of laying hens, but the presence of this steroid in the fowl was not established conclusively until Hertelendy et al. (1965) isolated crystalline material from hen’s urine, methylated this material and purified it and obtained the pure crystalline 3-methyl ether of estradiol-17β, in which form the steroid was characterized.
In the course of studies on the 24-hour urinary excretion of steroid estrogens by the hen (Common et al., 1965) as well as in connection with the isolation of estradiol-17β (Hertelendy et al., 1965), it was recognized that values for estradiol-17β obtained by Brown’s method (1955) were unreliable because of the presence of interfering Kober-positive material.
