Summary:Objective To investigate the effect of regulator of ribosome synthesis 1 (RRS1) on the proliferation and metastasis abilities of human breast cancer cells. Methods Western Blot was used to measure the protein expression level of RRS1 in human breast cancer cell lines (MDA-MB-231, MDA-MB-468, BT549, and MCF-7) and normal human breast epithelial cells. MDA-MB-468 cells were infected with sh-RRS1 lentivirus (sh-RRS1 group) and negative-control lentivirus (Con group), and MDA-MB-468 cells without infection were established as Blank group. Lentiviral infection efficiency was observed under a fluorescence microscope for the Con group and the sh-RRS1 group, and quantitative real-time PCR and Western blot were used to measure the mRNA and protein expression levels of RRS1 in cells; CCK-8 assay was used to observe the effect of RRS1 on the viability of MDA-MB-468 cells; scratch assay, Transwell assay, and invasion assay were used to observe the effect of RRS1 on the invasion and migration abilities of MDA-MB-468 cells
