鸭源NDV激活鸡和鸭先天性免疫应答的比较研究

Summary: | 鸡(Gallus gallus)ε 鸭(Anas platyrhynchos)是新城疫病毒(Newcastle disease virus,NDV)的主要宿主,但对于NDV感染的反应差异显著,先天性免疫应答在其中起重要作用。为了比较鸭源NDV激活鸡和鸭先天性免疫应答的不同,阐明先天性免疫应答在宿主致病性差异产生中的作用,以SD03(鸭源NDV)对鸡和鸭的致病性比较研究为基础,利用荧光定量PCR对不同时间点外周血中模式识别受体(pattern recognition receptors,PRRs)、细胞因子、抗病毒蛋白和主要组织相容性复合体分子(major histocompatibility complex.MHC)mRNA的表达量进行检测和比较。结果表明,SD03在鸡脾脏中的复制效率高于鸭;免疫基因中Toll样受体7(toll-like receptor 7,TLR-7)、白介素1β(interleukin 1β,IL-1β)、IL-2、IL-6、IL-8、干扰素β(interferon β,IFN-β)、MHCⅠ和MHCⅡ在鸡和鸭外周血中的表达模式不同且表达量变化趋势差异显著;除IL-2外,所有检测基因在鸡外周血中的表达量明显高于鸭。研究结果在机体水平上展示了NDV诱导鸡和鸭先天性免疫反应的不同,并分析了其在致病性差异产生中的作用,为全面理解NDV对不同禽类的致病机制提供了数据支持。 |
