SSc患者外周血外泌体通过microRNA-21介导成纤维细胞活化

Objective To explore the levels of microRNA-21-5p (miR-21-5p) in circulating exosome respectively isolated from systemic sclerosis (SSc) patients and healthy volunteers. The effects of exosome on skin fibroblasts were also evaluated. Methods Specific kits were used for circulating exosome isolation and RNA extraction. Levels of miR-21-5p were detected by qRT-PCR. CCK-8 was used to evaluate proliferation ability. Western blots were used to evaluate the protein levels of collagen I , collagen Ⅲ and a-smooth muscle actin (α-SMA). Results The levels of miR-21-5p were much higher in circulating exosome from SSc patients than that in healthy volunteers. Compared to healthy control, exosome from SSe patients could significantly elevated protein levels of collagen I , Ⅲ and α-SMA in skin fibroblasts, which could be partly blocked by miR-21-Sp transfection.
