Summary:目的研究丁胱亚磺酰亚胺(buthionine sulfoximine,BSO)联合放射线对食管癌细胞株TE-1放射增敏的影响。方法四甲基偶氮唑蓝比色实验(MTT法)观察BSO及放射线对细胞增殖的抑制作用及BSO对食管癌细胞株TE-1的放射增敏作用。流式细胞法观察BSO、放射线对细胞凋亡及细胞周期的影响。反转录聚合酶链反应(reverse transcription polymerase chine reaction,RT-PCR)、Western blot法观察BSO对锰超氧化物歧化酶(manganese superoxide dismutase,MnSOD)mRNA及蛋白表达的影响。结果 BSO能抑制食管癌TE-1细胞的增殖,具有明显的量效和时效关系;BSO对食管癌TE-1细胞有明显的放射增敏作用;BSO与放射线联合作用于TE-1细胞后,G2/M期细胞比例及凋亡率明显增加,MnSOD mRNA及蛋白表达下降。结论 BSO可能通过影响TE-1细胞周期,降低MnSOD mR-NA及蛋白的表达,从而抑制食管癌细胞增殖、诱导凋亡来增强放射线对食管癌细胞的杀伤作用。
