Summary:We investigated the positive effects of Undaria pinnatifida Sporophyll (USP) on lipid accumulation and oxidative toxicity in Caenorhabditis elegans, when obesity was induced by a high-glucose diet (HGD). As a result of the acute toxicity test, we confirmed that the concentration of UPS extract that did not affect survival was 62.5-500 μg/mL. UPS extract reduced total lipids in worms with obesity induced by a HGD. UPS showed a tolerance against oxidative and thermal stress and also protective effects on reactive oxygen species (ROS) generation in worms fed with HGD. In addition, UPS contributed to increasing the nuclear expression of daf-16. UPS extract was not effective in DPPH radical scavenging but increased the activity of SOD and GSH, the antioxidant enzymes in C. elegans. Therefore, these results suggest that UPS extract increases antioxidant enzyme activity through daf-16 regulation at the molecular level, exhibiting a protective effect against metabolic stress.
