Summary: | This study evaluated the antioxidant activity and immunomodulatory properties of Locusta migratoria hot water extract (LME). 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric reducing/antioxidant power (FRAP) assays demonstrated that the antioxidant capacity and reducing power of LME increase in a concentration-dependent manner. When RAW 264.7 cells, a type of macrophage that plays a crucial role in immunity, were treated with LME, no significant differences in cell viability were observed. However, the secretion of cytokines was significantly enhanced in the LME-treated group, as were nitric oxide (NO) secretion and inducible nitric oxide synthase (iNOS) /cyclooxygenase-2 (COX-2) expression levels. Additionally, it was confirmed that LME enhances immune activity through the phosphorylation of mitogen-activated protein kinases (MAPKs) and nuclear translocation of nuclear factor kappa B (NF-κB). These results suggest that LME may enhance the immune function of macrophages. This study highlights the potential of LME as a functional material for enhancing immune activity.
