Summary:Purpose: A mixture of Centella asiatica extract and Portulaca oleracea extracts (FSG-CA) was prepared in a 1:1 ratio. There have been no studies on the health benefits of this mixture to date. This in vitro study aimed to investigate the efficacy of FSG-CA in increasing the growth in height. Methods: HepG2 hepatoma cells were treated with various concentrations (0, 1, 5, 10, or 20 ㎍/mL) of FSG-CA. In HepG2 cells, the protein and mRNA expressions of insulin-like growth factor-1 (IGF-1) and IGF-binding protein-3 (IGFBP-3)- were measured by enzyme-linked immunosorbent assay (ELISA) and quantitative reverse-transcription polymerase chain reaction (RT-PCR), respectively. MC3T3-E1 osteoblastic cells were incubated in an osteoblast differentiation-inducing medium and treated with various concentrations (0, 25, 50, 100, or 200 ㎍/mL) of FSG-CA. In MC3T3-E1 cells, alkaline phosphatase (ALP) activity was assayed. Mineralization was determined by staining the cells with alizarin red S. In addition, the mRNA expressions of collagen type I alpha 1 chain (COL1A1), and bone morphogenetic protein-2 (BMP-2) were measured by quantitative RT-PCR. Results: In HepG2 cells, FSG-CA increased the production of IGF-1 and IGFBP-3. The mRNA expressions of IGF-1 and IGFBP-3 were increased in the cells treated with FSG-CA. In MC3T3-E1 cells, FSG-CA increased ALP activity, an early-stage marker of osteoblast differentiation, and mineralization, a late-stage marker of osteoblast differentiation. In addition, the mRNA expressions of COL1A1 and BMP-2 were increased in the cells treated with FSG-CA. Conclusion: These results demonstrate that FSG-CA can increase the expression of IGF-1 and IGFBP-3, growth regulators secreted by the liver, in HepG2 cells and osteoblast activity, a key action in longitudinal bone growth, in MC3T3-E1 cells. This suggests the potential of FSG-CA as a functional food ingredient for promoting growth in height. 본 연구에서는 병풀추출물과 쇠비름추출물을 1:1 비율로 혼합한 복합물 (FSG-CA)의 키 성장 효능을 탐색하고자 FSG-CA가 간세포주인 HepG2 세포에서 IGF-1과 IGFBP-3 생성과 조골세포주인 MC3T3-E1 세포에서 조골활성에 미치는 영향을 조사하였다. FSG-CA는 HepG2 세포에서 IGF-1과 IGFBP-3 발현을 유의적으로 증가시켰다. MC3T3-E1 세포에서 FSG-CA는 ALP 활성, COL1A1 mRNA 발현, 무기질 침착, 및 BMP-2 mRNA 발현을 유의적으로 증가시켰다. 이 결과는 in vitro 수준에서 FSG-CA가 간에서 분비되는 성장 조절인자인 IGF-1과 IGFBP-3의 발현 증가와 뼈의 형성에 기여하는 조골세포의 조골활성 증가를 나타내며, 이는 FSG-CA가 키 성장 증진 기능성 원료 개발의 후보물질로서의 가능성을 제시한다.
