Summary:The in vitro activity of temafloxacin (TMFX), a new fluoroquinolone, was evaluated against anaerobic bacteria, and the inducibility of Clostridium difficile overgrowth in murine cecum by 5-day administration of temafloxacin was also studied. The antimicrobial activity of ciprofloxacin, ofloxacin and norfloxacin were compared with that of TMFX. TMFX showed a broad spectrum, and was the most active against 45 reference strains (44 species) tested, although some species of genus Lactobacillus had low susceptibility to this compound. Against all recent clinical isolates tested, TMFX was the most active among antimicrobial agents used; 90% of Bacteroides fragilisisolates were inhibited at 3.13 μg/ml of the compound. TMFX showed strong bactericidal activity against B.fragilis. A 5-day administration of TMFX did not provoke the overgrowth of C. difficile in murine cecum.
