Summary: | It has been suggested that leukotrienes (LTs) play very important roles in the pathophysiology of anaphylactic shock and bronchial asthma. In the present investigation we studied the effect of indomethacin on leukotriene-induced contractile responses in guinea pig trachea and lung parenchyma strips in order to elucidate the contraction mechanism of LTs. 1) Dose-response curves of LTC4, LTD4 and LTE4 in guinea pig trachea strips shifted upward with continuous infusion of indomethacin solution, and this shift became more predominant by increasing the dose of indomethacin. 2) Dose-response curves of LTC4, LTD4 and LTE4 in guinea pig parenchyma strips shifted downward with continuous infusion of indomethacin solution, and this shift became more predominant by increasing the dose of indomethacin. The above results suggest that LTs may contract guinea pig lung parenchyma strips by producing a lung parenchyma contracting substance like thromboxane A2(TxA2). However, LTs probably contract guinea pig trachea strips directly without any production of bronchoconstrictors such as TxA2. |
