Summary:The effects of topically applied 5-fluorouracil (5-FU) ointment on hairless mice have been studied. Five per cent 5-FU ointment was applied once daily to the dorsal surface of the mice, totaling one, two or seven applications. Pigmentation and skin hypertrophy following changes such as erosion or ulcer have occurred in all the five mice which received two and seven applications of the drug, respectively. Acanthosis, the appearance of active (DOPA positive) melanocytes in the epidermis and dermal cyst wall, and a moderate increase in the dermal melanocytes were seen in the pigment area. Pseudocarcinomatous hyperplasia and a high increase in the dermal melanocytes were seen in the hypertrophic skin. It is suggested that pigmentation of the hairless mice skin induced by 5-FU is due to boththe appearance of the epidermal melanocytes and the increase in the dermal melanocytes, although pigmentation-inducing mechanism of the drug is not clear.
