Summary:INTRODUCTION and AIM: Intrauterine hyperglycemia environment during gestational diabetes mellitus may induce abnormal fetal development and metabolic phenotypes in adulthood. We explored the effect of palmitoleate as a functional lipid on skeletal muscle differentiation using myoblast cell under a high glucose environment. METHODS: L6 rat skeletal myoblast cultured and differentiated to myotube under 100mg/dL glucose (control) or 450mg/dL glucose (HG) condition. We examined the effect of palmitoleate against myoblast differentiation monitoring differentiation markers (pax3, myf5, myoD, myogenin, and MRF4) by RT-PCR. Phospho-Akt was detected by western blotting and cellular reactive oxygen species (ROS) was assessed by CellROX®. RESULTS: The gene expression of myogenic regulatory factor (MRF) and the level of the Phospho-Akt were significantly higher in HG than control with differentiation in accordance with increment of ROS production. Palmitoleate decreased ROS production mediated by HG, which in turn decreased MRF expression and phospho-Akt. CONCLUSION: We showed that palmitoleate suppressed myogenic differentiation in hyperglycemic conditions. In particular, trans-palmitoleate had a beneficial effect on suppressing the excessive differentiation induced by ROS.
