Abstract Ritipenem acoxil (RIPM-AC) is a new oral penem antibiotic. We investigated the pharmacokinetics of RIPM-AC and cefotiam hexetil (CTM-HE) by the cross-over method. The pharmacokinetics of RIPM-AC and CTM-HE, each orally administrated at a dose of 200 mg, were compared in healthy male volunteers. The Cmax values of ritipenem (RIPM) and cefotiam (CTM) were 2.09μgg/ml and 2.85μg/ml, respectively. The T1/2 of RIPM and CTM were 0.56 h and 0.71 h, and the AUC0-12 values were 2.10 μg·h/ml and 4.99 μg·h/ml. The mean urinary recovery rates of RIPM and CTM up to 12 h after a single oral administration were 11.3% and 38.9%.

The mean urinary recovery rates of RIPM and CTM up to 12 h after a single oral administration were 11.3% and 38.9%. 健常成人男子ボランティアを対象として, 新規経口ペネム系抗生物質であるritipenem acoxil (RIPM-AC) の体内動態について検討した。なお, クロスオーバー法でcefotiam hexetil (CTMHE) の体内動態についても検討した。健常成人男子ボランティアにRIPM-AC, CTM-HEともに200mg錠を単回投与し, クロスオーバー法で各薬剤の血漿中濃度, および尿中排浬を検討した。最高血漿中濃度 (Cmax) は, ritipenem (RIPM) では2.09μg/ml, cefotiam (CTM) では2.85μg/mlを示した。また, RIPM, CTMの血漿中濃度半減期 (T1/2) は, それぞれ0.56h, 0.71h, 12時間までの血漿中濃度曲線下面積 (AUC0-12) はそれぞれ2.10μg・h/ml, 4.99μg・h/mlであった。浩性体の累積尿中排泄率は12時間までで, RIPM 11.3%, CTM38.9%であった。
