SERUM AND SPUTUM CONCENTRATIONS AND CLINICAL RESULTS OF RU 28965 IN RESPIRATORY TRACT INFECTION

1. Serum and sputum concentrations of RU 28965 Serum and sputum concentrations of RU 28965 were measured in chronic respiratory tract infection. RU 28965 was given orally in a single dose of 300 mg before breakfast. A microbioassay was performed by the thin layer cup method using Micrococcus luteus ATCC 9341. The peak serum concentration of RU 28965 was 5.05μg/ml at 3h after a single administration of 300 mg. Peak sputum concentration was 0.48μEg/ml at 6h. 2. Clinical results RU 28965 was clinically evaluated in 11 patients with respiratory tract infection: five males and six females aged 24-67 years. RU 28965 was given orally to 2 patients in a daily dose of 200 mg, 2 in a daily dose of 300 mg and 7 in a daily dose of 400 mg in two divided portions. The duration of administration was 7 days in eight patients, 10 days in two and 18 days in one patient. A total of six strains (H. influenzae, 5 and S. aureus, 1) were identified in sputum before administration. All strains were eradicated. The clinical efficacy rate was 90.9%(10/11): excellent 5 patients, good 5 and fair 1. There were no side-effects, but 5% eosinophils were observed in one case. From the above results, we conclude that RU 28965 is an effective, safe and useful new macrolide.
