Summary:A new spectrophotometric method is proposed for the determination of cyclamate. It is based on solvent extraction of the complex formed between Methylene Blue hydrochloride and sodium cyclamate. The color intensity of the complex in the organic layer (660mμ) is proportional to the amount of cyclamate in the aqueous solution. The calibration curve is obtained as follows. Varying amounts of standard sodium cyclamate solution (10μg/ml, 010ml) are added to 4ml of Clark-Lubs buffer (pH 7.0) and 10ml of Methylene Blue solution (1.0×10-3M). It is extracted by shaking with 10ml of chloroform, and the absorbancy of organic layer is mearured at 660mμ against reagent blank. A linear relationship is obtained in the range of 110μg/ml of sodium cyclamate in the aqueous solution. Special references are given to the effects of some organic compounds including amino acids and sweetening agents. The proposed method was applied to the analysis of commercial "drink-drugs" after the separation of cyclamate by QAE-Sephadex chromatography. The recovery of added sodium cyclamate was very satisfactory.
