Summary:Tetraodontid juveniles (n = 54) were collected from Shibetsu Bay, Hokkaido, Japan, on August 30, 2022. Since morphological identification to species was difficult, DNA barcoding was performed. The nucleotide sequences of mitochondrial 16S rDNA (16S), cytochrome c oxidase subunit I (COI), cytochrome b (Cyt b), and the control region (CR) of these juveniles were determined. Database homology search and phylogenetic analysis using COI, Cyt b and CR sequences unambiguously determined these juveniles as the purple puffer Takifugu porphyreus, whereas those using 16S sequence could not diagnostically identify these juveniles. In recent years, the catch of T. porphyreus in Hokkaido has increased rapidly, but information on the juveniles from this area is scarce. Future research is needed to explore the possibility that earlier life stages of T. porphyreus may exist in the vicinity of Shibetsu Bay.
