Summary:Objective: Obesity and acute coronary syndrome (ACS) are common health problems of recent years. There are many candidate genes related to the genetic infrastructure of ACS and obesity. This study aimed to investigate the association of Leptin glutamine to arginine substitution (Gln223Arg), Resistin-420 Cytosine/Guanine (C/G), and proliferator-activated receptor-gamma (PPAR-[gamma]) proline to alanine substitution (Pro12Ala) polymorphisms in obese patients with ACS in the Turkish population. Methods: Fifty obese patients concurrently diagnosed with ACS and 42 healthy controls were included in this study. These polymorphisms were analyzed using the polymerase chain reaction-restriction fragment length polymorphism and agarose gel electrophoresis methods. Results: The PPAR-[gamma] Pro12Ala polymorphism Pro/Ala genotype (p=0.001) was found to be higher in the obese patients with ACS, while the (Proline/Proline) genotype (p=0.001) was significantly higher in the control group. The GC genotype (p=0.045) distribution of Resistin-420 C/G was found to be significantly higher in the controls compared to the patient group. Conclusion: Our study presents new findings that the PPAR-[gamma] Pro12Ala polymorphism Pro/Ala genotype is a risk factor for ACS in obese individuals, whereas Resistin-420 C/G polymorphism GC genotype and PPAR-[gamma] Pro12Ala polymorphism Pro/Pro genotype may be protective factors for ACS in obese individuals. Keywords: Acute coronary syndrome, leptin, resistin, PPAR-[gamma], gene polymorphism, PCR-RFLP Amac: Obezite ve akut koroner sendrom (AKS) son yillarin yaygin saglik problemlerindendir. AKS ve obezitenin genetik altyapisi ile ilgili bir cok aday gen bildirilmektedir. Bu calismanin amaci, Turk populasyonundaki AKS'li obez hastalarda Leptin glutamin-arginin substitution (Gln223Arg), Rezistin -420 Sitozin/Guanin (C/G) ve proliferator ile aktive edilen reseptor gama (PPAR-[gamma]) prolin-alanin substitution (Pro12Ala) polimorfizmlerinin etkilerini incelemektir. Yontemler: Calismaya AKS tanisi konmus 50 obez hasta ve 42 saglikli kontrol dahil edilmistir. Polimorfizmler polimeraz zincir reaksiyonu-restriksiyon parca uzunluk polimorfizmi (PZR-RFLP) ve agaroz jel elektroforezi metotlari kullanilarak analiz edildi. Bulgular: PPAR-[gamma] Pro12Ala polimorfizmi icin Pro/Ala (p=0,001) genotipi tasima orani hasta grubunda kontrol grubuna gore, Proline/Proline (Pro/Pro) genotipi (p=0,001) tasima orani ise hasta grubuna oranla kontrol grubunda anlamli duzeyde daha yuksektir. Rezistin -420 C/G polimorfizminin GC genotip (p=0,045) dagilimi ise hasta grubu ile kiyaslandiginda kontrol grubunda anlamli olarak yuksek bulundu. Sonuc: Calismamiz, obez bireylerde AKS icin PPAR-[gamma] Pro12Ala polimorfizminin Pro/Ala genotipinin bir risk faktoru, Rezistin -420 C/G polimorfizminin GC genotipinin ve PPAR-[gamma] Pro12Ala polimorfizminin Pro/Pro genotipinin ise koruyucu bir faktor olabilecegine dair yeni bulgular saglamaktadir.
