Summary:Background. No study is available with Salacia oblonga, an Indian medicinal plant, regarding its starch blocking activity. We, therefore undertook preliminary studies to confirm our hypothesis. Objectives. To study the effect of S. oblonga extract on alpha-amylase enzyme in-vitro, against starch induced postprandial glycemia in animals and healthy humans (acute clinical studies). Design. (1) In-vitro inhibitory effect of S. oblonga extract on alpha amylase enzyme. (2) In-vivo effect on postprandial glycemia in starch loaded albino Wistar rats and (3) acute clinical investigations, on postprandial glycemia in healthy subjects (starch meal). Results. Study 1 showed significant in-vitro alpha-amylase inhibitory effect (25-57%; p<0.05). Study 2 demonstrated significant reduction (34%; p<0.05) in postprandial glycemia in animals. In study 3, significant decrease (35%; p<0.05) was noticed in postprandial hyperglycemia in healthy subjects. Conclusion. These studies suggest that S. oblonga extract can be developed as an effective medicinal food and herbal remedy in preventing and managing diabetes.
