Summary:Several methods for lipid isolation and fatty acid analysis in two different strains of Saccharomyces cerevisiae were compared. As the most effective procedure the extraction with tetrachlormethane-diethyl ether-petrol ether mixture with an acid digestion step followed by fatty acid analysis by capillary GC was found. Other methods underestimated both the lipid content and the content of fatty acids.
