Summary:The genus Scutellaria L. which belongs to the family Lamiaceae, encompasses about 350 species [1]. 16 species of Scutellaria grow wild in the flora of Turkey [2]. Antitumor, antiangiogenic, hepatoprotective, antioxidant, anticonvulsant and neuroprotective effects of several Scutellaria species have been reported. The chemical composition of Scutellaria species have also been studied since 1889 and over 295 phytoconstituents have been isolated [1]. Although many HPLC studies have been published reporting the determination of several flavonoids [3], there is no HPLC work focusing on the phenlyethanoid profile of the Scutellaria species. In this study we have developed a simple, sensitive, precise, accurate and a validated reversed phase high performance liquid chromatographic method for the simultaneous determination of six phenylethanoid glycosides (calceolarioside D, neocalceolarioside D, verbascoside, isoverbascoside, leucoseptoside A, martinoside) in MeOH extract of the aerial parts of S. hastifolia and S. velenovskyi . The method involved the use of Zorbax® XDB C 18 column (4.6 × 150 mm, 3.5 µm) at 25 °C with the mixture of MeCN and H 2 O gradient (15 – 45% MeCN in 35 min.), constant flow rate 0.8 ml/min) as a mobile phase and detection at 330nm. Calibration plots were linear in the range of 3 µg/mL-120 µg/mL for calceolarioside D, neocalceolarioside D, verbascoside, isoverbascoside and martinoside, 1 µg/mL-120 µg/mL for leucoseptoside A. The amounts of above phytoconstituents varied among the species. This method can be used for the determination of phenylethanoid glycosides which constitute one of the major chemical classess of the genus Scutellaria .
