响应面法优化养殖大黄鱼蛋白质提取工艺

目的优化养殖大黄鱼蛋白质提取的工艺条件。方法通过筛选试验,以样品提取液中总蛋白的SDS-PAGE电泳图谱的条带数和清晰度为指标,得到养殖大黄鱼蛋白质的最适提取液。采用Plackett-Burman(PB)试验和最陡爬坡试验对提取过程中的影响因素进行评估,然后以提取液pH、提取液浓度和液固比为自变量,以样品提取液中的总蛋白含量为响应值,利用响应面分析法研究各变量及其交互作用对总蛋白含量的影响。结果选择2%SDS溶液为最佳提取液,得到养殖大黄鱼蛋白质提取的最佳工艺条件如下:提取液为3.50 g/100 mL的SDS溶液,pH值为8.3,液固比为17.5:1,提取温度为25℃,提取时间为2 h。在此...
