丹参注射液对高糖诱导的大鼠腹膜间皮细胞的过氧化损伤及E钙黏素和α平滑肌肌动蛋白表达的影响

Summary:目的研究丹参注射液对高糖作用下的大鼠腹膜间皮细胞过氧化损伤及E钙黏素和α平滑肌肌动蛋白(α-SMA)表达的影响。方法体外培养大鼠原代腹膜间皮细胞,第3代细胞经无血清培养基同步培养24 h后分为正常对照组、高糖诱导组(25 mg/mL葡萄糖)、甘露醇对照组(25 mg/mL甘露醇)和丹参干预组(15 mg/mL丹参注射液+25 mg/mL葡萄糖),测定各组细胞内丙二醛(MDA)和活性氧(ROS)含量及超氧化物歧化酶(SOD)活性;采用RT-PCR技术和Western blotting方法检测各组细胞内E钙黏素和α-SMA的mRNA和蛋白表达。结果与正常对照组比较,高糖诱导组细胞内ROS和MDA含量增高,而SOD活性降低,E钙黏素mRNA和蛋白的表达减少,α-SMA mRNA和蛋白的表达增加,组间比较差异均有统计学意义(P〈0.05)。与高糖诱导组比较,丹参干预组细胞内ROS和MDA含量减少,SOD活性增强,E钙黏素mRNA和蛋白的表达增加,而α-SMA mRNA和蛋白的表达减少,组间比较差异均有统计学意义(P〈0.05)。结论丹参注射液可明显减轻高糖诱导的腹膜间皮细胞的过氧化损伤,其对细胞内E钙黏素和α-SMA表达的调控作用可能与延缓腹膜纤维化作用有关。
