乙醇对心肌梗死大鼠冠脉血管新生的影响

目的 探讨乙醇对大鼠急性心肌梗死后冠脉侧支循环形成的影响及其机制.方法 取雄性SD大鼠随机分成4组:假手术组,空白对照组,小剂量乙醇组(50%乙醇,0.5 g· kg-1·d-1)及大剂量乙醇组(50%乙醇,5g·kg-1·d-1).制作心肌梗死模型,每组各8只,于术后第3天处死,留取梗死边缘区存活心肌,采用免疫组织化学SP法检测梗死边缘区心肌内微血管密度(microvascular density,MVD),ELISA法测定血管内皮生长因子(vascular endothelial growth factor,VEGF)及内皮抑素水平.通过方差分析研究各组间MVD及VEGF、内皮抑素表达水平...

Summary:目的 探讨乙醇对大鼠急性心肌梗死后冠脉侧支循环形成的影响及其机制.方法 取雄性SD大鼠随机分成4组:假手术组,空白对照组,小剂量乙醇组(50%乙醇,0.5 g· kg-1·d-1)及大剂量乙醇组(50%乙醇,5g·kg-1·d-1).制作心肌梗死模型,每组各8只,于术后第3天处死,留取梗死边缘区存活心肌,采用免疫组织化学SP法检测梗死边缘区心肌内微血管密度(microvascular density,MVD),ELISA法测定血管内皮生长因子(vascular endothelial growth factor,VEGF)及内皮抑素水平.通过方差分析研究各组间MVD及VEGF、内皮抑素表达水平有无差异.结果 与假手术组相比,空白对照组MVD明显增加(P<0.05),VEGF、内皮抑素水平明显升高(P<0.05);小剂量乙醇组MVD较空白对照组明显增加(P<0.01),VEGF明显升高(P<0.05),内皮抑素浓度明显降低(P<0.05);而大剂量乙醇组MVD则明显减少(P<0.01),VEGF水平与空白对照组相比差异无统计学意义,而内皮抑素明显升高(P<0.05).结论 乙醇能够改变心肌梗死后缺血心肌内VEGF和内皮抑素的表达,少量乙醇摄入可以通过增加VEGF的表达促进缺血心肌内的血管新生过程,而大量乙醇摄入则提高内皮抑素的表达而抑制缺血心肌的血管新生过程.
