重组嗜酸乳杆菌S层蛋白拮抗H9N2禽流感病毒对树突状细胞侵袭的研究

Summary:[目的]乳酸杆菌S层蛋白可结合树突状细胞特异性细胞间黏附分子3捕获的非整合蛋白(DC-SIGN)受体调节树突状细胞的成熟和分化,禽流感病毒同样可以通过与DC-SIGN的结合促进病毒在人源树突状细胞内的复制。为了探究S层蛋白是否竞争性与DC-SIGN结合,进而拮抗禽流感病毒侵袭树突状细胞,本试验构建表达嗜酸乳杆菌ATCC4356 S层蛋白的重组大肠杆菌,并探究重组S层蛋白抑制H9N2禽流感病毒侵袭树突状细胞的影响。[方法]以嗜酸乳杆菌ATCC4356的基因组为模板扩增S层蛋白基因并携带GST标签,构建重组质粒p GEX-4t-2-S并转化E.coli Rosetta-gami2(DE3),18℃低温诱导重组S层蛋白的表达,提取S层蛋白后经亲和层析进一步纯化;用纯化的重组S层蛋白提前作用于小鼠和鸡骨髓源树突状细胞,随后感染H9N2禽流感病毒,检测S层蛋白对禽流感病毒感染的拮抗作用。[结果]电泳结果显示成功扩增出1 295 bp的带有同源臂的嗜酸乳杆菌ATCC4356 S层蛋白基因;酶切得到2条带,分别为载体骨架4 951 bp,目的基因1 263 bp,表示成功构建p GEX-4t-2-S质粒,并且测序结果正确;电泳结果显示转化子中带有S层蛋白的基因,表明构建的质粒成功转入到DE3中并成功表达;电泳结果显示在相对分子质量70×103处有1条带,灰度分析表明亲和层析纯化后得到了纯度为95%的重组S层蛋白,产量为每1011CFU大肠杆菌11.9 mg;流式细胞检测结果证明S层蛋白能够使入侵小鼠和鸡树突状细胞中的H9N2病毒量减少25%~30%,实时荧光定量PCR结果表明HA、NA、NP和PB1基因的相对表达水平均达20%以上。[结论]重组大肠杆菌Rosetta-gami2(DE3)表达的S层蛋白能够有效地拮抗H9N2亚型禽流感病毒对小鼠和鸡树突状细胞的侵袭,为预防禽流感疫病提供新思路。
Bibliography:Objectives] It is reported that surface layer(S-layer) protein of Lactobacillus acidophilus binds to the DC-specific ICAM- 3-grabbing nonintegrin (DC-SIGN)receptor and regulates the maturation and differentiation of dendritic cells. In addition, avian influ- enza virus can be also combined with DC-SIGN to promote the replication of virus in dendritic cells. In order to explore whether S-layer protein could bind to DC-SIGN competitively and inhibit the invasion of avian influenza virus into dendritic cells, this study constructed recombined Escherichia coli( E. coli)which expressed S-layer protein of L. acidophilus ATCC4356, and explored its ability to inhibit the invasion of H9N2 avian influenza virus( AIV)into dendritic cells. [ Methods] The gene of S-layer protein was amplified from the genome of L. acidophilus ATCC4356 by PCR and followed with GST tag to construct the recombinant plasmid pGEX-4t-2-S.
